Before you can fire a ranged weapon, it needs to be both equipped and loaded. Equipping the weapon is the easy part; just go to your inventory screen and drag the weapon to the ranged weapon slot in the lower-right corner.
Ranged weapons have their own equipment slot, so you don't need to unequip your melee weapon; you can have both at the same time.
You get one ranged weapon slot for every hand that you have; rifles and bows require two hands, while pistols only need one. If you have multiple ranged weapons equipped, they'll alternate fire with each ranged attack you make unless you have the Akimbo skill, in which case they'll fire simultaneously.
Weapons start unloaded, so you need to reload a newly-equipped weapon by pressing the R key. Bows use arrows, and firearms use bullets or shells. Ammo is loaded directly from your inventory; it doesn't itself need to be equipped.
As long as a ranged weapon has at least one shot available, you can make a ranged attack by pressing the F key (for Fire). You can click on the object or creature you'd like to shoot, or use the arrow keys or numpad to cycle through available targets, then press spacebar to confirm your choice.
You can cancel your attack by right-clicking or pressing the Escape key.
The lower-left section of your equipment screen includes a slot for thrown weapons. You can have one object in this slot at a time, but if you have any identical objects in your inventory they will automatically reload when thrown.
Throwing items works the same as firing a ranged weapon; just press T instead of F to start the attack.
